The taxonomic identification of archaeological bones depends on the integrity of the pieces, which sometimes does not occur in sites highly impacted by urbanization. The objective of the research was to elaborate an experimental model using molecular biology techniques to assist in identifying fish species possibly present in the sambaqui da Prainha, in the center of the city of Rio de Janeiro, and of a concretion of the sambaqui de Camboinhas in the Oceanic Region of Niteroi/RJ. Universal molecular targets for fish were used in the experimental model and archaeological material. We could not recover fish DNA from the archaeological material, but this does not diminishes the found results. The experimental model has shown promise and can be improved and used in future zooarchaeological research.
